Long Acting Depot Formulation: History, Mechanism, its Manufacturing and Market Review

Abstract


Patient compliance is a very important factor in the treatment of any disease. Specially in
treatment of chronic diseases such as cancer, arthritis, psychosis patient need to take medicine
for longer time. In such cases long-acting depot injection became excellent option for treatment.
This drug delivery involves drug release over a period of several weeks and months. These depot
formulations can be prepared by using biodegradable polymer Poly (lactide-co-glycolide)
PLGA. Various methods are widely used for preparation of drug containing PLGA
microspheres such as solvent evaporation and solvent extraction method, spray drying method,
double emulsion method, phase separation coacervation technique. Depot injections can be
given through intramuscular and subcutaneous route which further releases drug over a period
of time.
