RP-HPLC Method for the Simultaneous Estimation of Hydrocodone and Pseudoephedrine in Pharmaceutical Dosage Form

Abstract


A simple, rapid, specific, accurate and precise high-performance liquid chromatographic method was developed for the estimation of Hydrocodone and Pseudoephedrine in pharmaceutical dosage forms. Phenomenex C18 (250×2.2 mm ID) 4.5 µm was the column used. Methanol: Water: TEA (Tri ethyl amine) in the ratio of 50:50:0.5% (v/v/v) was the optimized mobile phase and pH was adjusted to 7.5 by using ortho-phosphoric acid. The flow rate was 1.0 ml/min and effluents were monitored at 243 nm. The method was validated for linearity, accuracy, precision, specificity and sensitivity. The retention time of Hydrocodone and Pseudoephedrine was found to be 2.817 and 3.792 min respectively. Limits of Detection (LOD) for Hydrocodone and Pseudoephedrine were found to be 1.92 and 0.78 µg/ml respectively and Limits of Quantitation (LOQ) for Hydrocodone and Pseudoephedrine were found to be 5.84 and 2.37 µg/ml respectively.
