Spectroscopic and Chromatographic Methods for Estimation of Voriconazole in Bulk and Pharmaceutical Dosage Forms

Abstract


Rapid, accurate and precise methods have been developed for estimation of voriconazole in bulk and pharmaceutical dosage forms using Ultraviolet (UV) spectroscopy and reverse phase high performance chromatography (RP-HPLC). The λmax selected in UV spectroscopy method was 255 nm and the calibration curve was linear in the concentration range of 2-50 µg/mL with correlation coefficient (r2)>0.99. In RP-HPLC method, the separation was carried out on Kromasil C18 (250 x 4.6mm, 5μm) column using methanol as mobile phase at flow rate of 1 mL/min. UV detector was used for detection of eluents set at 256 nm. The drug was showing linear response in the concentration range of 2-40 µg/mL. Both the methods were validated as per ICH guidelines and the results were found within acceptance limits. The methods were successfully applied for estimation of voriconazole in tablet dosage forms.
