Formulation and Characterization of Liquisolid Tablet of Skeletal Muscle Relaxants and Anti-Inflammatory Drugs

Baclofen is a skeletal muscle relaxants and nimesulide is a Non- steroidal anti- inflammatory drug (NSAID). Baclofen is used in the treatment of muscle spasm, arthritis and spondylitis. NSAID’s are preferred to reduce the symptoms caused by muscle spasm i.e., severe pain and fever. Hence combination of drug is used to minimise the frequent use of the drug. The main objective of the present work was to enhance solubility and dissolution rate by formulating and developing liquisolid system. To reduce the frequency of administration and patient compliance, liquisolid tablets were prepared by developing the mathematical model with different carrier and coating material and non-volatile solvents viz. Avicel PH 102, Aerosil, PEG 400 and PG. After fixing the concentration of drug and solvent, the drug release were carried for desired time. Combination of baclofen and nimesulide in the form of conventional tablet were prepared and drug release study was compared with liquisolid tablet. The drug release rate for liquisolid tablet was 98.52±0.062% for baclofen and 69±0.4% for nimesulide in 60 min and for conventional tablet 50.68% for baclofen and 39.68% for nimesulide in 60 min. The prepared tablets were evaluated for other parameters like general appearance, drug content uniformity, hardness, friability and in vitro disintegration studies. The best fit model for all the formulations was Hixson- Crowell model and release of drug mechanism was found to be non- Fickian. All the twelve formulations were subjected to stability studies at room temperature (25±2˚C) for a period of 8 weeks and found that there was no change in drug content.
