Quantitative Determination of Isomeric Components of essential oils by RP-HPLC-PDA in multi-herbal tablets

Essential oils are lipophilic and highly volatile secondary plant metabolites, principally mono- and sesquiterpenes. The widespread usage and applications in biopharmaceutics and cosmoceutics of these components drives their quality assessments criteria to be established. Gas chromatography is one of the predominantly used technique for analyzing volatile oils. Although Gas Chromatography employs estimation designs operated at elevated temperatures leading for plausible structural alterations of thermo labile compounds. Hence, a novel gradient HPLC-PDA method was developed to separate and quantify essential oils such as phenylpropene (eugenol), terpenoid (carvone) along with isomers such as phenyl propanoids (anethole and estragole) and monoterpenes citral (neral and geraniol). Separation was achieved on C18 ACE Generix (250mm × 4.6 mm) column with trifloroacetic acid (TFA) 0.1% and acetonitrile as mobile phase. The developed method was validated as per ICH guidelines Q2R1, and can be used as alternative for gas chromatography of essential oils analysis. This method is suitable for routine quality control and also to evaluate the quality of essential oils in formulation.
