Formulation and Evaluation of Gastro retentive Effervescent Floating Tablets of Famotidine: Effect of Formulation Variables and In Vitro In Vivo Evaluation

Gastro retentive effervescent floating tablets of famotidine were developed in an attempt to sustain the drug release as well as to prolong the presence of dosage form in the stomach or upper small intestine until drug is completely released in desired period of time from the formulation with special emphasis to the effect and concentration of different polymers on drug release and floatation. Tablets were prepared by direct compression method and additives used were Sodium bicarbonate as gas generating agent, Di calcium phosphate and Micro crystalline cellulose as diluents, Magnesium stearate (2%) and talc (1%) as anti adherent and lubricant in the preparation of tablets. The prepared tablets were evaluated for Pre- compression parameters like Angle of repose, Bulk density and tapped density, Compressibility index, Hausner’s ratio etc and Post-compression parameters like in vitro buoyancy studies, Swelling index, Drug content estimation, In vitro dissolution studies, Compatibility studies by FTIR spectral analysis etc. Based on the desired tablet properties for floating tablets as well as in vitro release characteristics best formulations were selected and subjected to in vivo radiographic studies in New Zealand white species rabbits.
