Interaction of Cardiotoxin with Calmodulin and its EF Hand Peptides

Calmodulin is a ubiquitous protein having EF hand peptide(helix-loop-helix sstructural domain), which is found in all the eukaryotic cells. Calmodulin functions as a multipurpose intracellular receptor, mediating much calcium regulated processes and undergoes conformational change upon binding to calcium. These activated calmodul in then bind to a number of different target proteins, including a family of calcium/calmodul in-dependent protein kinases which then phosphorylatedserine’s or threnodies on other proteins. Cardiotoxin, snake toxins are known to act through the various cellular pathways. Many target proteins are known to bind calmodulin and exert their function therefore, this study has been designed in such a way to study the interaction of cardiotoxin three with calmodulin. Calmodulin was isolated from bovine brain, tissue was subjected to homogenisation followed by hydrophobic interaction chromatography. Stains—all, a carbocyanine dye was used in this study to record circular dichroism spectra. There was change in circular dichroism spectra when cardiotoxin was added to calmodulin-stains all complex. This decrease in J band on concomitant addition of cardiotoxin to calmodulin–satins all complex which show that there is interaction between Calmodulin and cardiotoxin. Cardiotoxin therefore by binding to calmodulin effect various biological process this needs to be studied in future.
